Planning to have a (72x18x30) 150G aquarium on top and a (48×18) 75G aquarium on the bottom. Will be finishing this off with plywood and a nice stain. In theory six 4×4’s should be plenty strong, right? Might add some cross bracing to the back to account for any sway. Interested in people’s thoughts.

Just checking. You going to add some support between top rim and bottom rim?

Your 4x4s are strong. However they are limited by the shear strength of screws at the moment. You have guide posts but now you need support posts.
Add the purple ones vertically and the dark purple horizontally.
